Is it possible to calculate the conjugate points when the lens is not symmetric? Can we design the lens to have two different focal length at the back and the front or even different focal lengths at different point on the lens?

Yes; an asymmetric lens by definition has focal points at different lengths at the back and front. The conjugate points would similarly be different on either side, but still easily calculable.
